    Background: Transforming growth factor-beta (TGF)-β inhibited epithelial cell proliferation and regulated a diverse array of cellular functions unrelated to cellular transformation. Today, the TGF-β superfamily is recognized as one of the largest families of secreted prototypic multifunctional peptides exerting an array of biological effects in a large variety of cell types. Objective: The objective of the study is to analyze the growth factor gene expression in buccal mucosal cell line. Materials and Methods: Cell line: TR146 was maintained at 5% CO2 in DMEM with FBS, penicillin, and streptomycin. Quantitative TGF-β gene expression: Total RNA was extracted from cell lysate and was reverse transcribed using reverse transcription system. Primer directed against the housekeeping genes ubiquitin C and glyceraldehyde-3-phosphate dehydrogenase was used as the endogenous control. TGF-β1 gene was amplified using the following primer. Result: TGF-β expression was prominent in TR146 cells as compared to the normal mucosal lysate. TGF-β inhibits proliferation and promotes both apoptosis and differentiation. Thus, it is evident that in buccal tumor cell line, TGF-β expression is very high. Conclusion: The study in human buccal tumor cell line samples has illustrated that disruption to TGF-β1 signaling promotes carcinogenesis.
