    Introduction: Irrigation is important for disinfection of the root canal system and participates in periapical repair, so their biocompatibility is of utmost importance to avoid tissue damage. The aim of this study was to evaluate the biocompatibility of sodium hypochlorite, ethylenediaminetetraacetic acid, and modified salt solution in the periapical tissue of rats according to the inflammatory reaction. Materials and Methods: The study was conducted on 18 male Wistar rats. Incision was made in the dorsal side on the mandibular region of the rats and the paste was placed in surgical holes drilled in the bone. After 7 days, the animals were euthanized and the specimens were subjected to histotechnical preparation. The hematoxylin and eosin-stained histological sections were analyzed by light microscopy. Scores were established according to the inflammatory process and statistically compared by the Tukey test (α = 5%). Results: The connective tissue was moderately inflamed at 7 days when contacting the experimental paste. The analysis of histological sections showed significant inflammatory reaction in the periapical region in all the three groups. The results were statistically significant. Conclusion: Within the limitations of the study, modified salt solution is used as alternative for sodium hypochlorite and ethylenediaminetetraacetic acid as an irrigating solution.
