Title: Study of the pharmacological activity of arginase II low molecular selective inhibitor on a monocrotaline-induced pulmonary hypertension model

    Introduction: Nitric oxide imbalance is one of the main pathogenesis elements of pulmonary hypertension (PH). Compound ZB49 as an arginase II selective inhibitor can be potentially used for the treatment of this disease. Materials and Methods: The work was performed on 40 adult male Wistar rats weighing 180–220 g. Simulation of monocrotaline (MCT) PH was performed on 20 animals using a subcutaneous injection of a spirit aqueous solution of MCT. After 4 weeks from the start of the experiment, animals were anesthetized and subjected to the measurement of the right and left ventricular pressure during sodium nitroprusside and acetylcholine injection with the subsequent calculation of the coefficient of endothelial dysfunction (CED). Results: The intragastric administration of an active pharmaceutical substation ZB49 at a dose of 5 mg/kg against the background of simulating MCT-induced PH revealed a statistically significant decrease in the CED, systolic pressure in the cavity of the right ventricle, average pressure in the cavity of the right ventricle, diastolic pressure in the cavity of the right ventricle, the maximum rate of contraction (dP/dt max), and the minimum rate of contraction (dP/dt min). At the same time, a statistically significant is positive effect of ZB49 on the gas composition of the blood. Conclusion: Compound ZB49 is able to improve cardiohemodynamics, blood gas composition, and decrease the mass of right ventricular of the heart in rats with PH.
