    Aim: Lactic acid bacteria (LAB) can be found in a variety of habitats, and several groups of these bacteria have been developed as probiotics. The objective of the present study was to isolate and identify LAB from the fresh milk of Toraja Belang Buffalo from South Sulawesi, Indonesia, and evaluate its potency as a probiotic candidate. Materials and Methods: Pathogenic bacteria used in the present study, EPEC ATCC 25922, Staphylococcus aureus 134-P, and Salmonella typhi ATCC 58105535, were obtained from the Microbiology Laboratory, Faculty of Medicine, Brawijaya University, Malang, Indonesia. Results: LAB were isolated by the pour plate technique using methicillin-resistant S. aureus-selective media containing 1% CaCO3 . LAB as a probiotic candidate were screened based on the hemolysis test on blood agar, the growth ability test at pH 3, 4, and 5, the bile resistance test at 0.3%, 0.5%, and 1%, and the ability to produce antimicrobial compounds against EPEC ATCC 25922 bacteria, S. aureus 134-P, and S. typhi ATCC 58105535. 14 isolates of LAB were obtained; of which, the K2B1 isolate was able to grow on pH three medium and 1% bile salt and to produce antimicrobial compounds against EPEC ATCC25922, S. aureus 134-P, and S. typhi. Conclusion: Based on the 16S rDNA sequence, the K2B1 isolate was Enterococcus faecalis, with 100% similarity to ATCC 19433. The K2B1 isolate can be developed as a probiotic candidate.
