    Objective: The present study aims to enhance the solubility and dissolution rate of telmisartan which is an angiotensin II receptor antagonist that can be used in the prevention and treatment of hypertension. Telmisartan belongs to BCS Class II moiety that has a low solubility in the biological fluids and, therefore, has a poor bioavailability after oral administration (about 42%) with a late onset of action. Materials and Methods: An effort was made to formulate a fast dissolving oral film containing telmisartan to improve its onset of action, therapeutic efficacy, patient compliance, and convenience of use. Various film-forming agents and polyhydric alcohols were used to prepare and evaluate the optimum composition of fast dissolving films. The films were prepared by solvent casting method using hydroxypropyl methylcellulose (HPMC) and polyvinyl alcohol as film-forming polymers, polyethylene glycol 400 as a plasticizer, tween80 as a surfactant, citric acid as a saliva-stimulating agent, and sorbitol as a sweetening agent. Solid state characterization was evaluated by differential scanning calorimeter (DSC) and Fourier-transform infrared (FTIR) spectroscopy. Results: DSC and FTIR spectroscopy analysis showed a complete transformation of telmisartan in the film from a crystalline state to an amorphous state. Different formulations of telmisartan films were prepared and evaluated for their physical characteristics that gave satisfactory results. The formulations were subjected to disintegration test and in vitro drug release studies, where formulation F1 was found to be the best formula among others which contain HPMC5 as a film-forming polymer that showed excellent characteristics such as disintegration time within 20 s and percentage drug release of about 100% within 7 min. Conclusions: Preparation of telmisartan as an oral film using a solvent casting method was considered as an excellent method to enhance the dissolution rate and the solubility of telmisartan.
