    Objective: The objective of the study was to carry out the acute toxicity studies and brain estimation of short peptide related to human histatin-8 in Alzheimer’s disease (AD). Materials and Methods: The behavioral, neurological, and autonomic profiles were determined by a rotation rod test and activity cage test. The histopathology of the brain structure of the rats was observed to determine the acute toxicity of the drug. The pharmacological evaluation was carried out by feeding aluminum chloride to the rat. The behavioral analysis was determined using the digital rotarod and activity cage to test the locomotor of the rats. The biochemical analysis was determined and identified by carried out the brain dissection and blood collection method. Results: There was no toxic effect shown in 2000 mg/kg of the peptide through the intraperitoneal route. The behavioral analysis data showed that some improvement in locomotor testing after injecting with the donepezil and human histatin-8. Biochemical analysis data showed that the donepezil could lead to renal and liver function impaired while human histatin-8 would not lead to renal failure due to the normal level of creatinine. For the pharmacological evaluation, the rat with the feeding of aluminum chloride showed poor locomotor movement in digital rotarod and cage activity. Conclusion: The dose of the drug should be maintained the same for the subsequent experiment as all the rats are still alive. Human histatin-8 are proven to be as the anti-AD which is significance in curing the AD. The preparation showed optimal pharmacokinetic characteristics in a rat model. During the injection of the human histatin-8, the rat model does not show the aggressive, depressant, and any serious adverse effect.
