    Background: Natural products have been used as potential inhibitors against carbohydrate‑hydrolyzing enzymes to treat hyperglycemia in type 2 diabetes (T2D) mellitus to minimize the toxicity and side effects of the inhibitors currently used to control hyperglycemia. Alpha‑amylase and α‑glucosidase inhibitors are used to achieve greater control over hyperglycemia in T2D mellitus. Objectives: The present study was indented to investigate the in vitro α‑amylase and α‑glucosidase inhibitory activities of iridoid glucoside isolated from the leaves of Vitex negundo. Results: Different concentrations (10, 20, 40, 80, and 100 μg/ml) of iridoid glucoside and acarbose were subjected individually to α‑amylase and α‑glucosidase inhibitory assay, and their absorbance was measured at 540 and 405 nm using a colorimeter. Iridoid glucoside showed appreciable α‑amylase and α‑glucosidase inhibitory potential in a dose‑dependent manner. The highest α‑amylase (67.22 ± 0.32) and α‑glucosidase (61.82 ± 0.30) inhibitory effects were found at 1000 μg/ml, respectively, and those inhibitory effects were comparable to that of acarbose ‑ used as a reference drug. Conclusion: These results indicated that iridoid glucoside exerted potent inhibitory effects against αamylase and α‑glucosidase which may be recommended as an oral antidiabetic drug.
