    Background: Allergic reactions induced by ingestion of foods containing sesame seeds are a well-recognized cause of severe food-induced anaphylaxis. Aim: The objective of the present study was to isolate, purify, and identify allergenic proteins of sesame seeds. Materials and Methods: Extract was obtained by the use of isoelectric precipitation, with 96.7% protein content of the product. Purified allergens were characterized by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), high-performance liquid chromatography, immunoblotting, and proteomics approach for amino acid sequence determination. Results: Seed proteins revealed that molecular sizes of all the reduced fractions were about 15–50 KDa by SDS-PAGE. Purified allergen was able to inhibit the immunoglobulin E binding to seeds extract in up to 90% of the responses. The identified tryptic peptide, AIFALPQAKIA of Black sesame and VQLAIHTLKSG of white sesame respectively, was commonly found in garden pea legumin. Conclusion: Majority of sesame proteins are minor allergens in regard to the prevalence and allergenic potency.
