Title: Antimicrobial and Phytochemical Screening inMyristica dactyloides Gaertn.

Antimicrobial and phytochemical screenings were identified in Myristica dactyloides Gaertn, The various solvent (petroleum ether, chloroform, and ethanol) extracts of the root bark, stem bark and leaves were tested by the agar well diffusion method against 12 human pathogenic bacteria and 6 human pathogenic fungus. Ethanol extract of root bark showed more activity of 16 – 21 mm to all the bacteria. As a whole, it has expressed 21 mm inhibition to four bacteria, where as 20 mm to one bacterium, Respectively 19 mm to two bacteria, 17 mm for four bacteria and 16 mm to one bacterium. In the case of fungi, the activities were 12 – 23 mm. plants do not express activity to fungi even when it expresses it is at the low profile. In the present study, the root bark exhibited a wide range of activities to both the bacteria and fungi. This plant has the potential for the development of drugs for various diseases. Petroleum ether extract could also be explored next to the ethanol extract.

Many modern medicines particularly contraceptives and microbicides are developed through the clues obtained from medicinal plants introduced by folk in traditional remedies worldwide. In this study, the spermicidal effects of the toxic latex of milk weed Calotropis procera- a well known abortifacient plant is investigated by in silico and in vitro methods. For studying in vitro spermcidal effects, doses of centrifuged fractions of aqueous latex were added to an equal amount of fresh human semen in 1:1 volumic ratio. Motility and viability of cells, effective concentration (EC 50) and sperm revival tests were carried out. The sperm immobilization effects of the aqueous extract appeared immediately but in a dose-dependent manner. Undiluted crude extract was the most effective and was able to immobilize human spermatozoa within five minutes whereas the diluted supernatant fraction was having non spermicidal effect. InSilico Ligand-Protein docking was attempted to better understand the binding mechanism of Calotropis sp with somatic and testis Angiotensin Converting Enzyme (ACE) receptors of human Renin Angiotensin System (RAS). Different amino acid residues that had hydrogen bonding with ACE receptors were identified. The in vitro screening study suggests that aqueous extract of latex of C.procera possess spermicidal activity on human sperm and the binding amino acid residues identified by in silico docking studies could be used in designing or developing future generation male contraceptives or microbicides.

In the present study, a new series of 2-Amino-3-(Substituted Benzylidinecarbohydrazide) -4,5,6,7-Tetrahydrobenzothiophenes (II a-i) were synthesized and screened for anti-inflammatory activity by in-vitro method. The structure of the compounds was characterized by IR spectral study and elemental analysis. Compound 2-amino-4,5,6,7-tetrahydrobenzothiophen-3-furylidinecarbo hydrazide (II i) showed significant anti-inflammatory activity. From this study, data revealed that carbohydrazide group is necessary for the potential activity.

Title: Phytochemical analysis and evaluation of antioxidant activities of hydro-ethanolic extract of Moringa oleifera Lam. Pods

The aim of present study is to investigate the phytochemical profile and antioxidant activities of pods of Moringa oleifera against free radicals using specific in vitro standard procedures. The antioxidant activity of the Hydro-ethanolic extract of pods of Moringa oleifera was evaluated by free radical scavenging activity using 1,1-diphenyl-2-picryl-hydrazil (DPPH), FRAP assay, reducing power assay, FTC, TBA and Non specific assay. The results of the assay were then compared with a natural antioxidant Quercitin, rutin, BHA and BHT. On the basis of present investigation the extract showed high significance (p<0.001) as compared to standards. The extract of the pods of Moringa oleifera is a good source of compounds with antioxidant properties while the extract also exhibited significant free radical scavenging activity, reducing power activity and Total antioxidant activity.

Title: A Validated RP HPLC Method for Simultaneous Determination of Propranolol hydrochloride and Alprazolam in Bulk and in Pharmaceutical formulations

The present work describes a new, simple, precise, and accurate RP-HPLC method for simultaneous estimation of propranolol hydrochloride and alprazolam in bulk and formulation. Chromatographic separation of the drugs was achieved on a Waters C18 column (250 × 4.6 mm, i.d. 5 ì) using acetonitrile: water (adjusted to pH 2.3 with ortho phosphoric acid) in the ratio of 60:40 v/v as mobile phase. Flow rate was 1.0 mL/min and the detection wavelength was 214 nm. The two drugs were satisfactorily resolved with retention time values 1.737 min and 3.810 min for propranolol and alprazolam, respectively. The method was validated in terms of accuracy, precision, linearity, and limit of detection, limit of quantitation, robustness, and ruggedness  as per ICH guidelines. Linearity was found to be in the range of 4 -30 µg/mL for propranolol hydrochloride and 0.05 – 0.375 mg/mL for alprazolam with significantly high value of correlation coefficient (r 2 = 0.9981 for propranolol hydrochloride and 0.9989 for alprazolam). The accuracy and reliability of the method was assessed by evaluation of precision (intra-day and inter-day precision % RSD was less than 2% for both alprazolam and propranolol hydrochloride), accuracy (100.01 % for propranolol hydrochloride and 99.84 % for alprazolam), and specificity, in accordance with ICH guidelines. The LOD and LOQ were found to be 0.4 ìg/mL and 1.2 ìg/mL for propranolol hydrochloride and 0.018 µg/mL and 0.05 µg/mL for alprazolam respectively. The proposed method can be used for the estimation of these drugs in combined dosage forms.

Ocimum sanctum is communally know as “The Queen of herbs ‘’ is the most sacred herb of India. Ocimum sanctum although also known as holy basil has been revered in India for over 5000 years, as a healing balm of body, mind and spirit and is known to bestow an amazing number of health benefit offers. Modern scientific research offers effective evidences that tulsi reduces stress, enhance stamina relives inflammation, lowers cholesterol, eliminates toxin, protects against radiation, gastric ulcer, improve digestion & provide a rich supply of antioxidants & other nutrients. Ocimum sanctum has reported effective in supporting the heart, blood vessels, liver & lungs and also regulates blood pressure and blood sugar. Tulsi has unique chemistry which is highly complex. The plant is reported to contain eugenol as a major chemical constituent which is a phenolic compound. Ocimum sanctum contain a number of beneficial compounds known as phytochemicals working together these compounds posses strong antioxidants, antibacterial, antioxidant, antiviral, adaptogenic and immune enhancing properties that promote general health and support the body’s natural defence against stress & disease. Sacred basil is now considered as a valuable source of unique natural products for development of medicine against various diseases. Present review compiles all the information/ scientific dataset aspects of pharmacological activity.

Considerable research works have been done on benzimidazole and their derivatives to evaluate their antibacterial, antiviral, anticancer, anticonvulsant, antihistaminic, anthelminitic and antidepressant activity and some of works have provided very promising results in antiasthamatic and antidiabetic activity. A wide variety of benzimidazole derivatives have been described for their chemotherapeutic importance. However there is a great need to study the various pharmacological actions derived from benzimidazole nucleus through research work.
