Title: Effects of Vitamin D3 on methotrexate-induced jejunum damage in rats by the utilization of immunohistochemical study

    Objectives: The main objectives of this study are to highlight the effects of fixed dose of Vitamin D3 at two different durations on the immunohistochemical expression of inducible nitric oxide synthase (iNOS) and caspase-3 induced by methotrexate (MTX) in the jejunum of rats. Materials and Methods: Forty-nine albino rats of both sexes, weighing 150–250 g were randomly divided into seven groups (seven animals each) as follows: Control 1 and control 2 (I.P. normal saline) daily for 14 and 21 days, respectively; Vitamin D3 groups (500 IU/rat/day) orally for 14 and 21 days, respectively; MTX a single dose (20 mg/kg, I.P) for 4 days; Vitamin D3 (500 IU/rat/day) once daily for 14 days and MTX (20 mg/kg I.P) which injected only at day 10; and Vitamin D3 (500 IU/rat/day) orally for 21 days and MTX (20 mg/kg I.P) which injected only at day 17; then, the jejunum was removed and used to investigate the expression of iNOS and caspase-3 by immunohistochemistry study. Results: The histochemical score of iNOS and caspase-3 protein in jejunum was significantly increased (P < 0.05) in MTX-treated group compared to control 1, control 2, and Vitamin D3 for 14 and 21 days; moreover, further increment in the histochemical score in Vitamin D3 administered for 14 or 21 days and single dose of MTX at day 10 or 17, respectively, compared to MTX-treated group (P < 0.05). Conclusion: The present study demonstrated that Vitamin D3 plays a synergistic role with MTX by increasing the expression of inducible NO and caspase-3 in rats’ jejunum.
