    Background: The existing pharmacological approaches for anxiety exhibited a slothful and a scanty reduction in anxiety may be due to its uncertain essential pathophysiology. Disturbances in cyclic adenosine monophosphate (cAMP) levels and its signaling pathways are attributed to anxiety. An alteration in the levels of cAMP levels would be a great approach for anxiety at the molecular level rather than affecting neurotransmitters directly. Increased expression of phosphorylated CREB (cyclic AMP response element-binding protein) may also be associated with anxiolytic activity. Phosphodiesterase inhibitors can elevate the cAMP levels by inhibiting its hydrolysis. Aim: The experiment is aimed to analyze the anxiolytic activity of cilostazol in two different doses alone and along with diazepam. Materials and Methods: Swiss albino rats and mice were used and randomly grouped into six groups with six animals in each group. They were treated with normal saline, diazepam 2 mg/kg, cilostazol 10 mg/kg, cilostazol 20 mg/kg, cilostazol 10 mg/kg + diazepam 2 mg/kg, and cilostazol 20 mg/kg + diazepam 2 mg/kg, respectively. Well-established and validated elevated plus maze and open-field models for anxiety were employed. Results: Cilostazol 20 mg/kg was exhibited significant difference over control and cilostazol 10 mg/kg in both anxiety models but not over standard drug diazepam 2 mg/kg. Among all groups, cilostazol 20 mg/kg + diazepam 2 mg/kg was exhibited grater statistically significant difference. Conclusion: The present preclinical experiment was established the anxiolytic and potentiating effect of cilostazol.
