Most regulators, including ICH, USFDA, Canadian Drug, and Health Agencies, stress the purity criteria for active pharmaceutical ingredients and the detection of impurities. The collection and assessment method of information establishing the biological security of an individual impurity is the classification of impurities, which demonstrates the needs and the context of impurity profiling of medicines in pharmaceutical research. Diverse chromatographic and spectroscopic methods are used for the detection of impurities, either alone or in conjunction with other techniques. Various methods are used for the identification and characterization of the Pharmaceutical substances. Identification of impurities is done by variety of chromatographic or spectroscopic techniques, either alone/in combination with other techniques. These are different methods for isolate and detecting & characterizing impurities in pharmaceuticals, such as, TLC, HPLC, and HPTLC. Conventional liquid chromatography (LC) is used extensively in the field of profiling of impurities in particular; the diverse applications contributed to the wide range of detectors, stationary stages, tolerance, and economic separation. The TLC is the most widely used separation system for impurity isolation and is one of the most versatile planar chromatographic processes, with low operating comfort and cost compared to HPLC. The improvement of chromatography is a popular process for the isolation of impurities. Headspace gas chromatography (GC) is one of the most common identification techniques for residual solvents. The introduction of hyphenated methods revolutionized the identification of impurities by not also distinguishing but also defining impurities structurally. Among the hyphenated methods, LC–mass spectrometry (LC–MS), LC–nuclear magnetic resonance (NMR), LC–NMR-MS, and GC–MS are the most advanced technologies for the characterization of drugs.
